The Research Group
The Genetic Mechanisms of Disease lab studies how genetic variation affects the normal functioning of the immune system and how this can go wrong in diseases like Crohn’s disease and ulcerative colitis. We use genomics, molecular biology, CRISPR-based methods and dynamic immunological assays to uncover disease mechanisms in primary human immune cells. Our goal is to not only discover the pathways that lead to immune-mediated disease, but to identify opportunities for the next generation of targeted treatments.
The Project
This role builds on our recent discovery of a transcription factor that functions as a master regulator of inflammatory responses in human macrophages and which is directly linked to multiple inflammatory diseases (https://www.biorxiv.org/content/10.1101/2023.05.05.539522v1). The project will explore the therapeutic potential of targeting this protein by selectively delivering inhibitors to macrophages via antibody-drug conjugates and related methods.
Research techniques used in this project will include primary immune cell isolation and culture, flow cytometry, biochemical activity assays, confocal microscopy, and other chemical biology techniques.

The ability to work in a team is essential.
Key experience and competencies
The post holder should embody and demonstrate our core Crick values: bold, imaginative, open, dynamic and collegial, in addition to the following:
Essential
PhD, or in the final stages of PhD completion, in a relevant biomedical science (including but not limited to: immunology, molecular biology, cell biology, biochemistry)
Experience in molecular biology, mammalian cell culture and flow cytometry
Knowledge of some / all of: human genetics, immunology and biochemistry.
Experience of working independently, designing experiments and adopting new techniques
Track record of scientific writing, evidenced by first author publications or submitted manuscripts in refereed journals
Ability to work independently and as part of a small team
Excellent organisational and communication skills
Desirable
Experience in chemical biology / synthesis
Experience in confocal microscopy
Experience in bioinformatics (preferably R/Bioconductor)
